Promenade -> ramparts -> bridge -> village -> clock tower -> clock room -> castle -> throne is a valid path, and the only entrances that require runes are getting to ramparts from promenade with vine rune (which you get from promenade) and finishing the run with homunculus (which you acquire in the same room).
